Asymbiotic Germination of Cymbidium ensifolium



Abstract
The study was conducted to get some informations on plantlet differentiation and growth of rhizome and seedlings through the several experiments, such as effects of peptone concns added to hyponex 3g/§¤ sucrose, agar, charcoal and coconut milk concns, pH value and dark and light culture periods with rhizome obtained by seeding of Cymbidium ensifolium in vitro.
Basal medium suitable for organogenesis and growth of rhizome and seedling was identified as a component, 3g/§¤ hyponex, 4g/§¤ peptone, 4.0% sucrose and 0.8% agar, this should be ajusted pH to 4.5.
Charcoal concns for organogenesis and growth of seedlings and rhizome was 1.0% and coconut milk concns for growth of rhizome, 15%.
It was noted that 2 weeks was the optimum dark period to stimulate organogenesis and growth of rhizome after transplanting.
